What it is

MOTS-c is a short peptide encoded not in the cell nucleus but in mitochondrial DNA — one of a small group of mitochondrial-derived peptides discovered in the last twenty years. It appears to act as a signal about the cell's energy state.

Composition

A sixteen-amino-acid peptide encoded within the mitochondrial 12S ribosomal RNA gene. What is sold online is a research chemical.

Proposed mechanism

It appears to act partly through the AMPK pathway — the same low-energy sensor that exercise and fasting activate — influencing glucose handling and fat metabolism. This is why it gets described as an exercise mimetic.

Claimed benefits

Improved insulin sensitivity, fat loss, better endurance, and slowed ageing.

What the evidence shows

Unproven· animal data only. The striking results are real and they are in mice: treated animals showed improved insulin sensitivity, resistance to diet-induced obesity and better running capacity. No controlled human trial has tested any of the physique or performance claims made for it.

The longevity angle rests on observation rather than intervention: MOTS-c levels fall with age, and certain genetic variants are more common in long-lived Japanese populations. That is an association in a legitimate line of ageing research, not a demonstrated effect of taking it.

Legal & regulatory status

Not a licensed medicine anywhere. As a non-approved substance it falls in a prohibited category under WADA rules at all times.

Known risks & unknowns

Essentially unknown in humans, because the human studies have not been done. That is the honest position: no established side-effect profile, no long-term data, and an unregulated product of unverifiable purity.
