Why it matters

The researchers conducted a systematic review and meta-analysis of six randomised controlled trials comparing tirzepatide with placebo in individuals with overweight or obesity who did not have diabetes mellitus. They pooled changes in percentage body weight, absolute body weight in kilograms, body mass index and waist circumference. On the safety side, they calculated relative risks for gastrointestinal side effects including nausea, vomiting, diarrhoea and constipation, as well as serious adverse events, serious gastrointestinal events, and discontinuation of treatment due to adverse events.

What they found

Tirzepatide reduced percentage body weight by a mean difference of -16.32% versus placebo (95% CI -18.35 to -14.29) and absolute weight by -13.95 kg (-18.83 to -9.07). Body mass index fell by -5.89 kg/m2 and waist circumference by -12.31 cm compared with placebo. Gastrointestinal side effects rose markedly: nausea (RR 3.11), vomiting (RR 5.94), diarrhoea (RR 2.92) and constipation (RR 2.85). Overall serious adverse events were not statistically higher than placebo (RR 0.93; 0.76-1.13), but serious gastrointestinal events (RR 3.07) and discontinuation due to adverse events (RR 2.29) were both significantly elevated.

This analysis extends the tirzepatide evidence base squarely into the non-diabetic population, confirming that the large weight reductions seen in earlier work hold up when trials are pooled and restricted to people without diabetes.
