Why it matters
L-theanine is a non-proteinogenic amino acid found in tea, and it has attracted sustained interest as a gentle option for sleep problems. Individual trials have tested it in different populations, at different doses and often in combination with other ingredients, which makes it hard to know whether the compound itself does anything for sleep. This review set out to pull together every randomised controlled trial of L-theanine supplementation and sleep, across all ages and health statuses, to establish what the evidence actually supports. For a supplement this widely discussed, a formal synthesis is overdue.
What they did
The researchers searched five electronic databases, APA PsycINFO, CINAHL, Medline, Scopus and Web of Science, plus the Cochrane Central Register of Controlled Trials, from inception until September 2024. They included randomised controlled trials investigating the effects of L-theanine supplementation on sleep quality in humans of any age or health status. Nineteen articles covering 897 participants were selected, and 18 of them contributed data to the meta-analysis. The pooled outcomes were subjective: how quickly people felt they fell asleep, how they functioned in the daytime, and how they rated their sleep overall.
What they found
L-theanine significantly improved subjective sleep onset latency, with a small pooled effect of 0.15 across 10 studies. Subjective daytime dysfunction improved by 0.33 across 9 studies, and overall subjective sleep quality score improved by 0.43 across 12 studies. All three effects reached statistical significance, but none is large, and all rest on how participants reported their own sleep rather than on objective recordings. The authors flag a structural weakness in the evidence: a lack of studies on pure L-theanine, as opposed to products combining it with other ingredients.
Where it fits
This is the most comprehensive pooling of L-theanine sleep trials to date, and it lands somewhere between the enthusiasm of supplement marketing and outright dismissal: the compound shows a consistent but modest signal on subjective sleep outcomes. The findings indicate potential use in managing sleep disturbances, but the field still cannot say what dose or duration works, and it cannot cleanly attribute effects to L-theanine alone when so many trials used blends. The authors call for future studies to settle dose, duration and effects in both healthy and clinical populations.
What it means for you
If you have seen L-theanine promoted as a sleep aid, this review is a reason to think the claim has some genuine trial support, and equally a reason to keep expectations modest. The improvements people reported were small, and because many trials tested combination products, the tea amino acid by itself may account for less than the pooled numbers suggest. The honest summary: a real signal, a small one, and a supplement whose optimal use nobody has yet worked out.
