Why it matters

Muscle protein synthesis is the process that repairs and builds muscle, and it briefly speeds up after a workout, a period often called the anabolic window. Whey protein is known to boost this response, but lifters have argued for years about when to take it and how much is worth taking. Most gym folklore says the shake belongs immediately after the session. The authors followed PRISMA guidelines and gathered 21 randomised controlled trials in healthy adults that combined whey protein supplementation with exercise. Fifteen studies had data suitable for pooling, while six were summarised qualitatively. Every eligible study measured either the myofibrillar fractional synthetic rate, a direct marker of muscle building, or the phosphorylation of proteins in the AKT/mTOR signalling pathway, all taken from muscle biopsy samples. That makes this evidence unusually direct: it comes from muscle tissue itself rather than from questionnaires or scales.

What they found

Whey plus exercise significantly raised the fractional synthetic rate, a large pooled effect. Timing mattered: synthesis rose 1.3 to 1.6 fold when whey was consumed immediately after exercise, but up to 2.5 fold when it was taken 45 minutes before multiple-set resistance exercise. The response also climbed with dose across the 10 to 60 g range tested. Signalling followed the same pattern, with phosphorylation of AKT, mTOR, 4E-BP1, p70S6K and rpS6 elevated 1 to 2 hours after exercise, and p70S6K and rpS6 falling back by 4 to 5 hours.

Where it fits

The post-workout shake is one of the most entrenched habits in training, and this review complicates it: the largest acute response came from protein taken before, not after, the session. That extends earlier work showing whey boosts the anabolic window, and suggests the window can be opened earlier than most people assume. What the review cannot show is whether a bigger acute synthetic response translates into more muscle over months of training, which is the outcome people actually care about. Trials tracking muscle growth directly under different timing schemes are the missing piece.

What it means for you

This is a reason to think that both the timing and the amount of protein around training shape the immediate muscle-building response, and that the pre-workout slot may be underrated. The authors conclude that 20 to 40 g of whey before multiple sets of resistance exercise may enhance synthesis and extend the anabolic window. It is acute biopsy evidence rather than long-term muscle measurement, so treat it as informative rather than settled. None of it changes the basic point that the response needs both the training and the protein to happen at all.
